Wonder Woman is a terrible character to begin with, so I think if anyone brave enough decides to tackle a project associated with her, than they need to do some major overhaul. As far as who plays her, I don't really care; just get an actress that can act, and not just look pretty and attempt striking poses. In fact, I'd probably say that a movie wouldn't be the best solution for her, in order to build her appeal. I'd probably go the tv show/series route.
As far as plot goes, I'd keep the focus in Themyscira, for the first couple of episodes, before having her placed in Earth. I'd want to give the viewer an opportunity to really see how an all-female, warrior-nation would function. I'd treat them almost like ants, in their brutal nature of overtaking small tribes surrounding them, with ongoing hints regarding their barbaric laws against males of ANY sort setting foot on lands they've claimed. I'd cut the lovey-dovey bull about how they are a kind nation, and turn them into a brutal Stratocracy who can handle out of state invasions well enough on their own. It would be bloody, and stomach curling, but it would offer insight to how these women are able to survive such attacks, by removing most sensitivity towards life that some of us carry with us today. I'd try my best to keep the soft-core nude scenes and cheap cop-outs about displaying their beauty out of the series all together. Just because they are an all-female nation, doesn't mean they follow all female stereotypes, nor does it have to submit to cheap 13-year old parlor tricks to keep the viewers attention. It's just not what they are about, and inputting such would destroy everything they stand for.
At any rate, the Queen and royalty of Themyscura are ones who do NOT directly partake in war, but they do handle the strategic placement of such. This being said, royalty are still raised by soldiers method, but they are expected to serve separately in strategic governing after reaching a certain age, rather than field combat. At this point, we see Diana(Wonder Woman) enter the scenery, constantly arguing with her mother, the Queen Hippolyta, that she doesn't want to be one of the strategic heads, or a future Queen, for that matter. She would rather be on the field with the rest of her peers battling, as she's more a fighter than a thinker(at least in her mind).
The contrast between her and her mother would play off pretty well, as they are of two different mindsets. The story develops and the viewer starts seeing the brutal nature of these women, and just how savage they are towards outsiders, especially males. It is said there are no males on the island of Themyscira, and any that are found shall be immediately be put to death once captured. Though indoctrinated with this belief, like all other daughters of the Amazon, Diana seems to be one of the few who sees this on loose moral grounds and is at constant odds with her mother regarding it. Eventually, Diana does some snooping and finds some very revealing developments regarding some well-hidden secrets among the elders and leaders of her nation, and begins questioning what it is she's defending.
As far as her mythology goes, there would be some winking to Wonder Woman fans regarding her comic lore. However, several things would change. For one, Diana would not be created from clay by her mother or Zeus, but it will play off as almost a religion or mythology to them as well. Her birth or background, as well as all other amazon's births, will be a major part of the plot, once it's revealed just how a nation of one sex can reproduce amidst homosexual relations among themselves.
Her name, "Wonder Woman", will be given to her as a nickname after she secretly enters a challenge among her peers to decide who shall be held as the new chiefs to head certain war parties, without her mother's or royalties consent, as royalty is banned from this competition. The winner was to be awarded the title after winning, by the Queen herself, but the Queen decided she wanted Diana to hand out the title, as it would begin an easement of succession to the people, by seeing the introduction of the new Queen mixing in political affairs with the people of Themyscira. After winning in combat against a fellow Amazon, her helmet is removed in anger by her opponent, and the Queen sees it's her daughter, Diana. The war chief title is taken away from Diana of course, and she is shamed in public by the Queen for her carelessness. Still ashamed, she slips out to personally apologize to those who's place she may have overstepped by entering and winning the competition. They accept the apology and accept her into the soldier's group for her granted talent for combat. While drinking, one of the soldiers makes a joke about her mother wondering where her daughter was to give her the title. She mimics Diana's mother by saying " I WONDER where the hell is that girl!", and plays with differing versions of it, while the others laugh at the mishap early in the day. Eventually, they dub Diana " Wonder Woman" due to it, and begin making up other stories in the drunk merry making, such as how she was made from clay by Zeus and other tall tales, to ease Diana's shame earlier in the day.
I have other ideas regarding different ways in which someone could handle her character and plot, but it's too long winded to even continue...basically, someone just needs to revamp her completely, and give her an actual reason to be worthwhile for girls/women to look up to.

You mention that these women can reproduce without men as a major plot plot. Good idea...IF you can make that work. There is a huge jump from 'good idea' to 'working idea', and that one is bigger than most. Considering that you also want to take the element of the gods and turn it into mythology and religion...and you'll have a harder time making this work. You HAVE to have a DAMNED good reason for it, one that people will accept as 'Wow, that blew my mind!'
And again, mythology is central to Wonder Woman's story and character. Remove it and it's not her. You'll have to include magic to explain why Themyscira has not been found and invaded by now (nations would take a dim view at some island that kills any man that goes there), especially since they have basically Iron-age tech. You also have to explain powers...because if WW doesn't have powers, it's pointless to do this at all.
Also keep in mind that the best Wonder Woman runs in the comics are based on the concept of her being 'The Paragon of Truth'. While a warrior, she is also a healer and diplomat. She sees things as they are and acts with a purity of purpose that rivals Superman. Make her all emo and emotionally scarred...and you got that hideous TV Kelley tried to make.

The plot could be kind of like that with Ares as the villain. also, kind of like the Thor movie, they could have some sci-fi elements to explain the origins of the people of Themyscira.
